We are encountering a few people where their holiday entitlement is reflecting incorrectly (accrual calculations seem to be wrong). Is this a known issue- is there something we need to change to have their entitlement show correctly? Alternatively, are we able to manually update this to reflect the correct entitlement?

There isn't a known issue with holiday entitlement calculation and because this situation involves specific employees for a specific organisation, it is best that you raise a support ticket for this.
This release note does contain some guidance on flexible holiday calculations, if that helps?
